چکیده

Bladder cancer is a disease of continuing public health significance. Some known risk factors for bladder cancer development include smoking, occupational exposure, genetic susceptibility, infectious diseases and radiation therapy. Smoking and occupational exposure have strongly implicated aromatic amines as being carcinogenic for the bladder, with 4-aminobiphenyl (4-ABP) being one of the most potent. 4-ABP is a major etiological agent of human bladder cancer, and its metabolites are able to form DNA adducts that may induce mutation and initiate bladder carcinogenesis. Binding characteristics and specificity of bladder cancer anti-DNA antibodies were analyzed by direct binding and inhibition enzyme-linked immunosorbent assay. The data show preferential binding of bladder cancer antibodies to 4-ABP-NOmodified DNA in comparison with native native DNA. A band shift assay further substantiated the enhanced recognition of 4-ABP-NO-modified DNA by anti-DNA antibodies. Cancer antibodies exhibited enhanced binding with the modified human DNA as compared to the native form. Lymphocyte DNA from cancer patients showed appreciable recognition of anti-4-ABP–NO-DNA IgG as compared to the DNA from healthy subjects. The 4-ABP-NOmodified DNA presents unique epitopes which may be one of the factors for the autoantibody induction in bladder cancer patients.

Genotoxic Effect of N-Hydroxy-4-Acetylaminobiphenyl on Human DNA: Implications in Bladder Cancer

BACKGROUND The interaction of environmental chemicals and their metabolites with biological macromolecules can result in cytotoxic and genotoxic effects. 4-Aminobiphenyl (4-ABP) and several other related arylamines have been shown to be causally involved in the induction of human urinary bladder cancers. The genotoxic and the carcinogenic effects of 4-ABP are exhibited only when it is metabolic...

Immunochemical quantitation of DNA adducts derived from the human bladder carcinogen 4-aminobiphenyl.

To assess target-tissue exposure to the human urinary bladder carcinogen 4-aminobiphenyl (ABP), we have developed a sensitive immunochemical method for measuring the major arylamine-DNA adduct formed, N-(guan-8-yl)-ABP (Gua-C8-ABP). High-affinity polyclonal antisera from rabbits immunized with N-(guanosin-8-yl)-ABP coupled to keyhole limpet hemocyanin were characterized and shown to have high s...

Impact of Cryopreservation Process on Viability, Nitric Oxide and DNA Apoptosis in Fertile Human Spermatozoa

Introduction: Infertility is a common phenomenon in modern societies. Today, use of assisted reproductive technologies (ARTs) for treatment of infertility is common, and cryopreservation is one of these technologies. Cryopreservation has impact on the function and percentage of fertility of human sperm. In this study, motility, viability, nitric oxide, and DNA apoptosis were assessed before and...

Carcinogen-DNA adducts in exfoliated urothelial cells: techniques for noninvasive human monitoring.

Detection of carcinogen-DNA adducts in DNA from exfoliated urothelial cells from animals and humans exposed to potential environmental carcinogens is described. In an animal model, 4-aminobiphenyl-DNA adducts were detected, and the shape of the dose-response curve was related to the levels of 4-aminobiphenyl-hemoglobin adducts. In a human study, five distinct adducts were two to nine times high...

A modified host cell reactivation assay to measure DNA repair capacity for removing 4-aminobiphenyl adducts: a pilot study of bladder cancer.

As DNA repair plays an important role in genetic susceptibility to bladder cancer, assessment of the DNA repair phenotype is critical for the molecular epidemiology of bladder cancer. In this study, we developed and applied an assay using the luciferase (luc) reporter gene in a host-cell reactivation assay to measure DNA repair capacity for DNA damage induced by 4-aminobiphenyl (4-ABP), a well-...
